The talented three-year-old was forced to dig deep with 60.5kg in the www.royalrandwick.com Handicap (1600m) before coming out on top in a thrilling battle with the Gai Waterhouse-trained Teardrop Rock.
"I think that was his best win," Farley said.
"He had a lot of weight and he had no favours in the run in the small field.
"He had to cart them all up, do it all himself, but Chris (O'Brien) knows the horse and he knows what he's got.
"Chris has always said he would like to see him get into a dogfight and he did that today."
Sincero will now be sent for a well-earned one month spell and Farley sees no reason to change the plans he has mapped out for the horse's next campaign.
He will target the Listed Gosford Guineas first-up before heading to the Hawkesbury Guineas and Scone Guineas.
His owners have rejected offers up to $1.1 million and have no intention to sell.
"That will do him the world of good being forced to chase like he did today," Farley said.
"He'll have a month off now and he'll know he has had that run today."
The Boxing Day victory took Sincero's record to six wins from nine starts and the son of Umatilla has been unbeaten in four runs this preparation.
O'Brien was happy to sit back in the field of five and keep the $1.45 favourite in clear galloping room as Tim Clark took Teardrop Rock ($8) to the front.
Teardrop Rock, who was also chasing her fourth straight win, kicked strongly and the pair settled down the fight out a driving finish.
O'Brien was always confident Sincero would pull out enough to win and he did so to get up by a short half-head.
Winning Glory ($8.50) was 3-1/2 lengths further back in third as Sincero smashed the class record in his first test over the mile.
Waterhouse was delighted with Teardrop Rock and said the filly was being aimed at the Tasmanian Guineas and Tasmanian Oaks.
