Cassidy was elated after receiving the call from trainer Tracey Bartley to partner Sniper's Bullet in Western Australia's premier weight-for-age race at Ascot on Saturday week.
The hoop has been sitting on 99 Group One winners since 2006 after booting County Tyrone home in the Sydney Cup.
"It's about time I raised my bat," Cassidy said of the milestone of 100 major winners.
The Kingston Town Classic (1800m) was formerly known as the Fruit 'n' Veg Stakes, a race Cassidy won on St. Clemens Belle in 1999.
"I love Perth racing and was pretty happy to get the call from Tracey this morning," Cassidy said.
"To go over there to ride such a nice horse who is going so impressively is something I'm really looking forward to."
Sniper's Bullet, who won the Stradbroke Handicap last year, finished a short head second to Gilded Venom in last Saturday's Group One Railway Stakes at Ascot.
Cassidy is one of an elite group of jockeys to have won the four major races on the Australian racing calendar - the Melbourne Cup, Cox Plate, Caulfield Cup and Golden Slipper.
The "Pumper" made the most of his only ride at Warwick Farm on Wednesday when he steered the classy Rainbow Styling to a slashing 5-1/2-length win in the Ashcroft Handicap (2100m).
